Methicillin is a narrow spectrum beta-lactam antibiotic. It is used in laboratory to test the antibiotic sensitivity of Staphylococcus aureus as compared to other penicillin that faces beta-lactam resistance.

Methicillin resistance is provided by the activation of a new bacterial penicillin binding protein, also known as mecA gene.

Methicillin inhibits the synthesis of the bacterial cell wall, where it inhibits the cross-linkage that occurs between the linear polymer peptidoglycan chains of the cell wall of gram-positive bacteria.
