Concept explainers
To explain: The use of Flow Cytometry in the diagnosis of leukemia.
Introduction: Flow Cytometry is an approach which sorts different cell type. The cell mixture is stained with particular dyes coupled with fluorescently labeled antibodies. The mixture of the cells is allowed to pass through a tiny liquid stream through a beam of laser light. The fluctuations in the intensity of light and the color of fluorescence are analyzed. Based on these observations each cell is sorted according to their DNA content, surface proteins, and morphology.
Explanation of Solution
In B-cell lymphoma, B-cells are known to express variations in their morphology as well as the expression of various surface receptors example CD19. CD19 is a significant cell surface receptor. It is expressed by all the B-cell population, at every stage of development.
If a human is expected to have B-cell lymphoma then flow- cytometry can be used as a diagnostic tool by doing the following experiments:
- Performing immunostaining of the cells with anti-CD19 antibody and one other fluorescent dye which absorb at a different wavelength.
- By determining the number of cells in the S(synthesis)/M (metaphase) of the cell cycle which are also CD19 positive
If a higher number of CD19 positive cells are present in the S (synthesis phase) and M (metaphase) of the cell cycle. then this observation shows that these cells are proliferating on a high rate and high number of B-cells indicates the condition of B-cell lymphoma.
Hence, the above observations are an indication of B-cell lymphoma condition.

- Which of the following is NOT a type of cell? A. ribosome B. haemocytoblast C. neutrophil D. phagocytearrow_forwardIf you run IHC and the antibody you chose binds to everything in a non-specific manner, what visual result would you see when looking at the tissue? A. No color change anywhere on the tissue B. A blank image on the microscope C. The entire tissue changing color (Brown staining) D. Half of the tissue changing colorarrow_forwardWhen vesicular stomatitis virus (VSV) is used to infect mice via footpad injection, viral particles are trapped in the draining lymph node (the popliteal lymph node) within 5 minutes of injection. These viral particles are then retained in the lymph node for many hours, where they can be visualized on cells that are interacting with B cells. The cells retaining the viral particles in the lymph node are not tissue-resident dendritic cells that have migrated to the lymph node with the virus, as this process takes much longer than 5 minutes. In which region of the lymph node would you expect to find the trapped viral particles and on which cells?arrow_forward
- Considering that antibodies are proteins that will be released from the cell, which types of cytoplasmic organelles are likely tobe found in high numbers in plasma cells? (See table 3.1 for a summary of organelles and their functions.)arrow_forwardResearchers can make monoclonal antibodies by immunizing a mouse with a molecule (or with a microorganism). The resulting antibody-mediated response produces a set of antibodies that recognize different parts of the molecule. The mouse's B cells are then harvested from its spleen and fused with cancerous B cells from a myeloma cell line. The resulting hybrid myeloma ("hybridoma") cells are cloned: Individual cells are grown in tissue culture as separate cell lines. Each cell line produces and secretes antibodies that recognize one part of the immunizing molecule. These antibodies are called monoclonal antibodies, and they can be purified and used for research or other purposes. Monoclonal antibodies are effective for passive immunization, but only in the immediate term. Antibodies produced by one's own immune system can last up to about six months in the bloodstream, but monoclonals delivered in passive immunization often last for less than a week. Why the difference?arrow_forwardCancer cells have a complex interaction with the cells of the immune system that are present in the stroma. The immune cells have the potential to destroy the tumor if it is recognized as aberrant tissue. Which one of the following statements correctly describes an interaction between cancer cells and the immune cells of the stroma? A. Tumors stimulate formation of new blood vessels—angiogenesis—promoting tumor survival. B. The tumor establishes an immunosuppressive environment by blocking white-cell activation. C. Tumors invoke an inflammatory reaction that blocks entry of immune cells into the stroma. D. Immune cells block tumor growth by providing signals that inhibit cancer cell proliferation.arrow_forward

- Why is a cell-mediated immune response more effective against intracellular microorganisms (such as viruses) than an antibody-mediated response? Select one: a. Cell-mediated (vs. antibody-mediated) response results in activation of many more immune cells, so it is more effective at destroying antigens. b. Intracellular microorganisms are too large for antibodies to be effective against. c. Cell-mediated response results in destruction of cells containing the intracellular microorganism; antibodies can not enter cells. d. Intracellular microorganisms are too small for antibodies to be effective against.arrow_forwardHenry showed his mother the bite mark that he just received from a large bug in his backyard as he was playing.
