Concept explainers
To review:
The way, by which an activated G protein such as Ras, amplify a response to ligand binding.
Introduction:
G-protein coupled receptor (GPCR) as a signaling pathway is the most common of all. G protein is a lipid-anchoring protein, which is made up of 300 amino acids. It has alpha (α), beta (β), and gamma (γ) subunits, and is heterotrimeric. Gα is linked with guanosine diphosphate (GDP) at one end and to the plasma membrane at another end. It is the largest subunit and has guanosine triphosphatase (GTPase) action. Gβ and Gγ are the other two subunits of G protein.
GPCR signaling pathway has receptor and effector molecules. Effector molecules are generally plasma membrane-bound enzymes like adenylate cyclase and guanylyl cyclase. These effector molecules activate secondary messengers, which in turn, activate the cytosolic enzymes.

- Briefly explain in your words the role of how Inside-out signaling of integrin occurs?arrow_forwardDoes adenylate cyclase become more active when the inhibitory (Gia) protein is mutated and increases its intrinsic GTPase activity?arrow_forwardContinuous exposure of a G protein-coupled receptor to its ligand leads to a phenomenon known as desensitization. Describe several molecule mechanisms for receptor desensitizationarrow_forward
- What are the most common sites on proteins that are phosphorylated during signal transduction cascades?arrow_forwardIs the term “G protein” just a shorter name for aG protein-coupled receptor?arrow_forwardThe ras protein is a mutated G protein that lacks GTPase activity. How does the absence of this activity affect the adenylyl cyclase pathway?arrow_forward
- RGS proteins accelerate the GAP activity of Gα subunits. What is the direct result of this GAP activity? - hydrolysis of GTP, resulting in a GDP-bound Gα - phosphorylation of GDP to GTP, resulting in a GTP-bound Gα - an exchange of GTP for GDP bound to Gα - an exchange of GDP for GTP bound to Gαarrow_forwardCan you please describe the role of G proteins in ligand-activated cellular signal transduction.arrow_forwardWhen you think about the primary structure of the mannose-6-P receptor, assuming that it is an integral membrane protein with one membrane-spanning domain and assuming that it has a ERSS, what are all of the different protein domains that must be present in this protein's primary structure to get it to where it needs to go and to carry out all of the functions/activities? (Draw a schematic of the protein structure/sequence and explain each region). What do you think would happen to a soluble protein that normally contains a KDEL tag (ER retrieval) if the tag was cleaved off (where would the protein end up)?arrow_forward
- Members of the integrin family are the major cell surface receptors for many ECM proteins such as fibronectin. In experimental studies, single integrin heterodimers have been found to assume more than one structural conformation. Describe the relationship between integrin structures and their activities.arrow_forwardBetween direct and indirect allosteric kinase inhibitors, which do you think requires a larger conformational change in response to binding of the allosteric modulator? Why?arrow_forwardSubstrate binding causes dimerization of tyrosine kinase receptors. Why is dimerization critical to activate of the receptors?arrow_forward
